kleisinの名称はギリシア語で閉鎖を意味するkleisimoに由来する。これは、V字型のSMC2量体のヘッドドメインに結合して「環を閉じる」働きが想定されているためである。kleisinファミリーに属するタンパク質の代表的なものとして、真核生物ではコンデンシン複合体のサブユニットCAP-HとCAP-H2や、コヒーシン複合体のサブユニットRad21/Scc1(減数分裂期ではRec8やRad21L)などがある。原核生物型コンデンシン複合体のサブユニットでは、枯草菌のScpAや大腸菌のMukFがこれに相当する。
kleisinの構造
kleisinのC末端はwinged helix構造をとり、SMCタンパク質のヘッドドメイン先端部("cap"領域)と結合する[2][3][4]。一方、kleisinのN末端はもう一方のSMCヘッド付近のコイルドコイル領域(”neck”領域)に結合する[5][6][4]。中央領域には、HEATリピートサブユニット(真核生物型コンデンシンやコヒーシンの場合)やkiteサブユニット(原核生物型コンデンシンやSMC5/6複合体の場合)が結合する。
